Primer set for detecting oncogenic mutations in single cells

The invention relates to a set of primers enabling sensitive detection of oncogenic mutations in DNA including single cell material. The primers are adapted for DNA amplified with Ampli1 WGA and for analysis using MALDI TOF mass spectrometry.

The panel detects key somatic mutations in TP53, PIK3CA, ERBB2, ESR1, CDH1 and AKT1 which allows confirmation of the cancerous origin of rare cells such as circulating tumor cells and supports rapid assessment of tumor heterogeneity.

The method provides high sensitivity, short analysis time and reliable results even from minimal amounts of DNA.

Application:

  • Identification of cancer origin of single cells,
  • Assessment of tumor heterogeneity and disease progression,
  • Screening of mutations in samples with very low DNA yield.
